Bombing hits sports fans in Afghanistan

KABUL, Afghanistan (AP) ? Afghan officials say several people have been killed and injured when a suicide bomber blew himself up in a crowd of sports fans watching a buzkashi game in northern Afghanistan.

Amanddin Quriashi, a senior official in the Imam Sahib district of Kunduz province, says the district police chief is among the wounded in Wednesday's bombing.

Kunduz province police chief Gen. Mohammad Khalil Andarabi says the father of the speaker of the Afghan parliament was among those killed in the attack.

The provincial police chief's Sayed Sarwar Hussaini has confirmed the suicide attack but says the number of casualties is still being determined.

The crowd was watching a game of buzkashi, a traditional Afghan sport akin to polo, where players on horseback use a headless goat carcass instead of a ball.

'Mad Men' sneak peek: More of Don, less Betty

The "Mad Men" creator and cast finally dropped some hints about what we can expect when the long-awaited sixth season finally debuts April 7.

"It's not what you think is coming," teases Jessica Par?, who plays Don Draper's second wife, Megan, in a behind-the-scenes featurette. "It's not coming in the way you thought it was coming. It's probably not coming at all. Unless you were expecting more great costumes and more big hair."

Big Betty, however, is no more. Guess that grapefruit diet paid off, because January Jones' alter ego is showing off a svelte figure in a new promo picture.

Although the still -- a formalwear shot featuring Don, Megan and a very mature-looking Sally Draper -- is in black and white, the new behind-the-scenes video captures the garish getups of the era.

Rich Sommer, who plays Harry Crane, sports a hideous brown shirt, brown and burnt orange striped tie, a brown and white checked blazer and giant -- you guessed it -- brown glasses.

"It's certainly one of the more exciting seasons," Sommer enthuses about the series' sixth installment. "There's a lot of stuff happening that would never have happened on the show three or four years ago."

Notoriously secretive showrunner Matthew Weiner coyly adds, "We left off in 1967 -- there is no year left between now and then that is not filled with upheaval."

Weiner also promises "a lot of insight into Don, a new dynamic, and people striving to grow up."

"There are definitely two sides to life," notes Hamm. "The part of life that is public and the part that is private. A little bit what we're trying to do is sort of pull back the curtain."

The curtain will finally open on "Mad Men's" sixth season Sunday, April 7 at 9 p.m. on AMC.

SEFFNER, Fla. (AP) ? Engineers worked gingerly Saturday to find out more about a slowly growing sinkhole that swallowed a Florida man in his bedroom, believing the entire house could eventually succumb to the unstable ground.

It could be days before officials decide whether they will attempt to recover Jeff Bush's body, and they were still trying Saturday to determine the extent of the sinkhole network and what kind of work might be safe. As the sinkhole grows, it may pose further risk to the subdivision and its homes.

Bush, 37, was in his bedroom Thursday night in Seffner ? a suburb of 8,000 people 15 miles east of downtown Tampa ? when the earth opened and took him and everything else in his room. Five others in the house escape unharmed.

On Saturday, the normally quiet neighborhood of concrete block homes painted in Florida pastels was jammed with cars as engineers, reporters, and curious onlookers came to the scene.

At the home next door to the Bushes, a family cried and organized boxes. Testing determined that their house also was compromised by the sinkhole, according to Hillsborough County Fire Rescue spokesman Ronnie Rivera. The family, which had evacuated Friday, was allowed to go inside for about a half-hour to gathering belongings.

Sisters Soliris and Elbairis Gonzalez, who live on the same street as Bushes, said rumors are swirling among neighbors, with people concerned for their safety.

"I've had nightmares," Soliris Gonzalez, 31, said. "In my dreams, I keep checking for cracks in the house."

They said the family has discussed where to go if forced to evacuate, and they've taken their important documents to a storage unit.

"The rest of it, this is material stuff, as long as our family is fine," Soliris Gonzalez said.

"You never know underneath the ground what's happening," added Elbairis Gonzalez, 30.

Experts say thousands of sinkholes erupt yearly in Florida because of the state's unique geography, though most are small and deaths rarely occur.

"There's hardly a place in Florida that's immune to sinkholes," said Sandy Nettles, who owns a geology consulting company in the Tampa area. "There's no way of ever predicting where a sinkhole is going to occur."

Most sinkholes are small, like one found Saturday morning in Largo, 35 miles away from Seffner. The Largo sinkhole, at about 10 feet long and several feet wide, is in a mall parking lot. Such discoveries are common throughout the year in Florida.

The state is prone because it sits on limestone, a porous rock that easily dissolves in water, with a layer of clay on top. The clay is thicker in some locations ? including the area where Bush became a victim ? making them even more prone to sinkholes.

Jonathan Arthur, the state geologist and director of the Florida Geological Survey, said other states sit atop limestone in a similar way, but Florida has additional factors ? extreme weather, development, aquifer pumping and construction ? that can cause sinkholes. "The conditions under which a sinkhole will form can be very rapid, or they can form slowly over time," he said.

But it remained unclear Saturday what, if anything, caused the Seffner sinkhole.

"The condition that caused that sinkhole could have started a million years ago," Nettles said.

Engineers had been testing in the area of the Bush house since 7 a.m. Saturday. By 10 a.m., officials moved media crews farther away so experts could test a home across the street.

Experts spent the previous day on the property, taking soil samples and running tests ? while acknowledging that the entire lot where Bush lay entombed was dangerous. On Saturday, officials were still not allowing anyone in the Bush home.

Jeremy Bush, who tried to rescue his brother when the earth opened, lay flowers and a stuffed lamb near the house Saturday morning and wept.

He said someone came to his home in the Tampa suburb of about 8,000 people a couple of months ago to check for sinkholes and other issues, apparently for insurance purposes, but found nothing wrong. State law requires home insurers to provide coverage against sinkholes.

"And a couple of months later, my brother dies. In a sinkhole," Bush said Friday.

The sinkhole, estimated at 20 feet across and 20 feet deep, caused the home's concrete floor to cave in around 11 p.m. Thursday as everyone in the Tampa-area house was turning in for the night. It gave way with a loud crash that sounded like a car hitting the house and brought Jeremy Bush running.

Engineers said they may have to demolish the small house, even though from the outside there appeared to be nothing wrong with the four-bedroom, concrete-wall structure, built in 1974.

Yahoo to shut down seven products, including Blackberry app

SAN FRANCISCO (Reuters) - Yahoo Inc is shutting down seven products, including its mobile app for Blackberry smartphones, as new Chief Executive Marissa Mayer takes a page from Google Inc's Playbook by eliminating unsuccessful products en masse.

The product shutdowns, which Yahoo announced on its official company blog on Friday, are part of what the company said are regular efforts to evaluate and review its product lineup.

"The most critical question we ask is whether the experience is truly a daily habit that still resonates for all of you today," wrote Jay Rossiter, Yahoo's executive vice president of Platforms.

The announcement represents Yahoo's second group shutdown of products since Mayer, a former Google executive, became CEO of the struggling Web portal in July. So-called "spring cleaning" announcements, in which multiple products are shut down, have become a regular feature at Google in recent years.

Mayer signaled the company would prune its line-up of mobile apps at an investor conference last month, noting that Yahoo would reduce the 60 to 75 disparate mobile apps it currently has to a more manageable 12 to 15 apps.

Yahoo said its app for Blackberry smartphones would no longer be available for download, or supported by Yahoo, as of April 1.

Yahoo also said that on April 1 it will stop supporting Yahoo Avatars - the cartoon-like digital characters that consumers create to depict them on Web services such as Yahoo instant messenger and Facebook. Consumers who want to continue using their avatar on Yahoo's online services must download the avatar and then re-upload the information to their personalized Yahoo profile.

The other Yahoo products set to be terminated include Yahoo App Search, Yahoo Sports IQ, Yahoo Clues, the Yahoo Message Boards website and the Yahoo Updates API.

Two new genes linked to amyotrophic lateral sclerosis (ALS) and related disorders

Mar. 3, 2013 ? A study led by St. Jude Children's Research Hospital has discovered mutations in two genes that lead to the death of nerve cells in amyotrophic lateral sclerosis (ALS), also known as Lou Gehrig's disease, and related degenerative diseases.

The same mutation occurred in both genes and led to the abnormal build-up of the proteins inside cells. These proteins play an essential role in normal RNA functioning and have also been linked to cancer, including the Ewing sarcoma, the second most common type of bone cancer in children and adolescents. The finding is the latest in a series of discoveries suggesting degenerative diseases and cancer may have common origins. RNA is the molecule that directs protein assembly based on instructions carried in DNA.

The study also adds to evidence that seemingly unrelated neurodegenerative diseases may involve similar defects in RNA metabolism. Researchers linked the problems to a specific region of the mutated proteins whose normal function was unclear. The study was published today in the advanced online edition of the scientific journal Nature.

"I hope this study helps to build the foundation for desperately needed treatments for ALS and perhaps a broad range of diseases caused by abnormal RNA metabolism," said J. Paul Taylor, M.D., Ph.D., an associate member of the St. Jude Department of Developmental Neurobiology and senior author of the study. Taylor and James Shorter, Ph.D., an assistant professor in the biochemistry and biophysics department at the University of Pennsylvania's Perelman School of Medicine, are the study's corresponding authors.

Each year approximately 5,600 people in the U.S. are found to have ALS. The disease is nearly always fatal, often within five years. Patients suffer muscle wasting and paralysis that affects their limbs and trunk as well as their ability to talk, swallow and breathe. There is no cure.

For this project, St. Jude sequenced just the portion of the genome called the exome, which carries instructions for making proteins. Researchers sequenced the exomes of two families affected by rare inherited degenerative disorders that target cells in the muscle, bone and brain. Neither family carried mutations previously tied to ALS or related diseases. The project built on the infrastructure developed by the St. Jude Children's Research Hospital -- Washington University Pediatric Cancer Genome Project, which played an important role in finding the mutations.

Researchers found the families carried a single, previously unknown mutation in a pair of RNA-binding proteins named hnRNPA2B1 and hnRNPA1. The proteins both bind RNA and help regulate its function. When researchers checked for the same mutations in 517 ALS patients they found hnRNPA1 protein mutated in two patients. One patient had the inherited form of ALS. The other ALS patient had no family history of the disease.

The new mutations occurred in a region of the proteins Taylor refers to as a prion-like domain because it has similarities with yeast proteins called prions. Prions are proteins that can alternate between shapes as needed for different functions. "Until recently we did not know these domains existed in humans and now we realize that hundreds of human proteins have them," Taylor said. "We're only beginning to understand their function in human cells."

Researchers showed the prion-like domains are responsible for the shape change that occurs when these proteins convert into slender threads called fibrils. The mutations accelerate fibril formation and recruit normal proteins to form fibrils. This phenomenon called propagation may explain how ALS and related diseases spread throughout the nervous system.

Taylor speculated that the normal function of prion-like domains is to assemble RNAs into temporary structures called granules, which are part of the cell's normal protein production machinery. Granules are normally short lived, and the RNA-binding proteins involved in their formation are recycled. But in cells with hnRNPA2B1 or hnRNPA1 mutation, RNA granules accumulated in the cytoplasm instead of being disassembled. "That's bad news for RNA regulation, which is bad news for those cells," Taylor said.

The study has several important implications, Taylor said. Recognition that the mutations adversely impact regulation of RNA could lead to targeted therapy to correct the problem. The mutation's location in the prion-like domain might also prove significant. Although the mutations in hnRNPA2B1 or hnRNPA1 appear to be rare, hundreds of other RNA-binding proteins have prion-like domains. Taylor said patients with unexplained neurodegenerative diseases may have mutations in these proteins.
